ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh komposisi Diclorophenoxy Acetic Acid (2,4-D) dan Benzil Amino Purin (BAP) terhadap pembentukan kalus eksplan pucuk nilam (Pogostemon cablin Benth) dengan pemotongan secara hotizontal. Penelitian ini menggunakan media Murashige & Skoog (MS) dengan kombinasi Diclorophenoxy Acetic Acid (2,4-D) dan Benzil Amino Purin (BAP). Penelitian ini dilakukan dengan 16 kali perlakukan (P0-P15) dan lima kali pengulangan (U1-U5). Parameter penelitian yang diamati pada penelitian ini berupa persentase pertumbuhan kalus, waktu kemunculan kalus (HST), berat kalus dan morfologi kalus. Hasil penelitian menunjukkan bahwa persentase pertumbuhan kalus yang diperoleh rata-rata sebesar 2,25% dengan waktu kemunculan kalus paling cepat adalah pada hari ke-16 pada perlakukan P1U5; P10U4; P12U2; P12U5; dan P13U2. Sementara berat basah kalus tertinggi diperoleh sebesar 0,19 gram pada perlakuan P8U2. Hasil juga menunjukkan bahwa kalus yang dihasilkan memiliki warna kuning dan krem, dengan tekstur kompak dan remah. Kata kunci : Pogostemon cablin Benth., pemotongan secara horizontal, kalus, 2,4-D, BAP ABSTRACT This study aims to determine the effect of the composition of 2,4-Diclorophenoxy Acetic Acid (2,4-D) and Benzyl Amino Purin (BAP) on the formation of patchouli (Pogostemon cablin Benth.) shoot explants by hotizontal cutting method. This research used Murashige & Skoog (MS) as growth medium. This research were conducted with 16 treatments (P0-P15) and five replication (U1-U5). The research parameters observed in this study were percentage of callus growth, time appearance of callus (day after planting, DAP), callus of weight and the morphology of callus. The results indicated that the percentage of callus growth obtained was by an average 2.25% with the time appearance of callus (day after planting, DAP) was 16 DAP at P1U5; P10U4; P12U2; P12U5; and P13U2. The highest weight of callus obtained was 0.19 grams at P8U2. The results also showed that the callus produced had a yellow and cream color, with a compact and crumb texture.
