ABSTRAK Ekstrak etanol daun pepaya (Carica papaya L.) diduga mempunyai efek hipoglikemia karena mengandung flavonoid, alkaloid, saponin dan tannin. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol daun pepaya terhadap kadar glukosa darah mencit yang diinduksi aloksan. Penelitian ini merupakan penelitian eksperimental dengan rancangan post test with control group design. Penelitian ini menggunakan mencit jantan (Mus musculus L.) galur Swiss-Webster dewasa sebanyak 25 ekor yang dibagi ke dalam 5 kelompok secara random. Kelompok K- (kontrol negatif) diberi air suling; K+ (kontrol positif) diberi glibenklamid dengan dosis 0,65 mg/kgBB/hari; P1, P2 dan P3 diberi ekstrak etanol daun pepaya dengan dosis 0,5 gr/kgBB/hari; 1 gr/kgBB/hari; dan 2 gr/kgBB/hari. Penelitian ini dilaksanakan selama 14 hari. Pemeriksaan kadar glukosa darah dilakukan pada hari ke-0, ke-3, ke-7 dan ke-14 menggunakan glukometer. Kadar glukosa darah pada hari ke-0 menunjukkan perbedaan yang signifikan antara kelompok K- dengan K+, P1, P2 dan P3. Pada hari ke-3 ditemukan perbedaan yang signifikan antar kelompok K- dengan K+ dan P1. Pada hari ke-7 tidak ditemukan perbedaan yang sifnifikan antara kelompok perlakuan. Sedangkan pada hari ke-14 didapatkan kelompok perlakuan K+, P1 dan P2 lebih tinggi kadar glukosa darahnya dibandingkan dengan kadar glukosa darah pada kelompok K- dan P3. Terdapat perbedaan kadar glukosa darah antar kelompok pada uji ANOVA dan tidak terdapat perbedaan yang signifikan antar kelompok K- dengan P3 yang ditunjukkan dengan uji Duncan. Ekstrak etanol daun pepaya yang diberikan pada mencit berpengaruh terhadap penurunan kadar glukosa darah. Kata kunci: Pepaya (Carica papaya L.), Kadar Gula Darah, Diabetes Mellitus. The study is aim to understand the effect administering ethanol extract of papaya’s leaf on blood glucose content of mouse that induced by aloksan. This study was experimental by posttest with control group design. It was use male mouse (Mus musculus L) the adult Swiss-Webster groove as many as 25 that divided into 5 groups randomly. K- Group (negative control) added distilled water; K+ (positive control) added glibenclamide by dose 0,65 mg/kgBW/day; 1 gr/kgBW/day; and 2 gr/kgBW/day. This study was conducted for 14 days. Examination for blood glucose content be done at 0, 3th, 7th, and 14th using glucometer. Blood glucose content in 0 day showed significant difference between group K- to K+, P1, P2 and P3. In 3th of day found the significant difference between K- Group to K+ and P1. In 7th of day there was not found significant difference among of treatment groups. While in 14th of day obtained groups of treatment K+, P1 and P2 were higher for the blood glucose content than K-Group and P3. There was difference of blood glucose content between among of groups in ANOVA test and there was no significant difference between K-Group to P3 that have shown by Duncan test.
