ABSTRAK Termo-halofilik merupakan bakteri yang mampu hidup pada suhu lingkungan ekstrem yaitu suhu tinggi dan kadar garam tinggi. Bakteri termo-halofilik dapat bertahan hidup pada kondisi lingkungan ekstrem karena mampu menghasilkan enzim termostabil. Pada penelitian ini bakteri termofilik berasal dari sumber air panas Pria Laot Sabang. Penelitian ini dilakukan dari bulan april sampai bulan juni 2014 di Laboratorium Biokimia, Laboratorium Mikrobiologi Fakultas MIPA dan Laboratorium Rekayasa Material Fakultas Teknik Universitas Syiah Kuala, Banda Aceh. Penelitian ini bertujuan untuk mengidentifikasi golongan isolat bakteri yang telah diisolasi dari sumber air panas Pria Laot Sabang secara fenotipik dengan mengamati morfologi koloni (bentuk, tepian, elevasi dan warna isolat), morfologi sel (pewarnaan Gram, anailisis SEM) dan aktivitas biokimia (motilitas, TSIA, MR-VP, LIA, Simmon Citrat, oksidase, katalase, methyl red, oksidatif dan fermentatif). Hasil penelitian menunjukkan bahwa ketiga isolat bakteri merupakan kelompok Gram negative dengan bentuk sel batang (basil) dan motil. Ketiga isolat secara umum memiliki morfolgi yang sama antara lain koloni berwarna krem, bentuk koloni buta, elevasi cembung dan tepian rata.berdasarkan hasil uji biokimia menunjukkan bahwa ketiga isolat bakteri merupakan kelompok family Bacilliceae, Genus Geobacillus dan jenis Geobacillus stearothermophillus. ABSTRAC Thermo-halophilic is a bactery survives in an ekstreme environtmental temperatures like high temperature and high salinity. Thermo-halophilic bacteria can survive in a ekstreme environtmental condition because it produces Thermostable enzymes. In this study Thermophilic well-born from Pria Laot Sabang Hot Springs. This study has started from April to June of 2014 in the Chemical laboratory, Microbiology Laboratory of MIPA faculty and Materials Laboratory of Engineering faculty, Syiah Kuala University. This study is to identify classes of bacteria that have been isolated from Pria Laot Sabang Hot Springs phenotypically by observing the colony morphology (type, edge, elevation, and colour isolated), cell morphology ( gram staining, SEM analysis) and biochemistry actifity (motility,TSIA,MR0VP,LIA, Simmon Citrate, Oxidase, Catalase, methyl red, Oxidative, and Fermentative). This Study showed that the three isolates of bacteria is a group of gram-negative with stem cells form and motile. The three isolates of bacteria generally have the same morphology as if crem colored-colonies, shape ball colonies, convex elevation, and flat ledges. Based on the biochemistry test result showed that the three isolates of bacteria are group of family Bacilliceae, Genus Geobacillus, and type of Geobacillus stearothermophillus.
