mencit yang diperoleh secara aspirasi dikultur selama 7 hari dalam inkubator co2 5% pada suhu 37 °c. parameter penelitian ini yaitu hasil kurva amplifikasi yang ditampilkan pada software cfx-96 biorad yang menunjukkan kesesuaian antara gen sel target dan primer sel tulang yang digunakan. primer yang digunakan adalah alkaline-phosphatase (alp). hasil penelitian menunjukkan bahwa ekstrak etanol bunga flamboyan dengan dosis yang digunakan mampu menginduksi diferensiasi sel punca menjadi sel tulang berupa osteoblas. kata kunci : bunga flamboyan (delonix regia (boj. ex hook.) raf.), sel punca mesenkimal, diferensiasi, sel tulang, real time pcr abstract stem cells technology has a great potential to cure degenerative diseases. the objective of this study was to identify genes that expressed by bone cell. it was a differentiation product of mesenchymal stem cell bone marrow in mice (mus musculus) induced by flamboyant d. regia (boj. ex hook.) raf. flower using real time pcr. this research was conducted at research laboratory of department of biology, faculty of mathemarics and natural sciences and faculty of veterinary medicine, syiah kuala university from june 2016 to february 2017. it prepared according to completely randomized design (crd) methode using five treatments and four replications. flamboyant d. regia flower extract was added to modifief dulbecco’s modified eagle’s medium (mdmem) with doses 0.0 mg/ml ( no addition extract), 0.6 mg/ml, 0.7 mg/ml, 0.8 mg/ml and 0.9 mg/ml. mice bone marrow cells which were obtained by aspiration were cultured for 7 days in 5% co2 incubator at 37 °c. parameter of this study was amplification curve which is presented by software cfx-96 biorad that showed conformity between gene target and alkaline-phosphatase (alp). the results showed that flamboyant flower extract with all doses used were able to induce the differentiation of msc into bone cells that is osteoblast. keyword : flamboyant (delonix regia (boj. ex hook.) raf.) flowers, mesenchyme stem cells, differentiation, bone cells, real time
